This product is a kit for DNA extraction from oral swabs, urogenital swabs, sputum, etc., developed by Taizhou Huaxia Ruitai Medical Instruments Co., Ltd. The BIOG DNA Swab Kit uses the latest high-quality imported ion membranes, and the lysate and eluent have been optimized several times to effectively separate DNA. Compared with other brands, the extracted DNA has higher yield, higher purity, and maximum removal of impurities such as proteins, pigments, and lipids. It can be directly applied to PCR, real-time PCR and various enzyme digestion tests. .



Bring your own: absolute ethanol, physiological saline, 1.5mL centrifuge tube.
Take out the supernatant and washing liquid as follows:
a) Precipitation: add 4.5 mL to 25.5 mL of absolute ethanol; add 9 mL to 51 mL of absolute ethanol.

b) Washing solution: add 9 mL of 21 mL of absolute ethanol; add 18 mL of 42 mL of absolute ethanol.

c) If there is precipitation in the prepared precipitation and washing solution, it can be dissolved at 37°C and shaken for later use.

Sample processing:
a) Swab: Add 800 μL of normal saline to a 1.5 mL centrifuge tube, put the sample collected by the swab in normal saline, rinse for 20 seconds to completely detach the cells, and squeeze the swab on the wall of the swab. liquid. Centrifuge tube. Centrifuge at 12,000 rpm for 5 minutes, discard 700 μL of supernatant, save 100 μL of supernatant, and mix for 15 seconds.

b) Sputum: add 4 times the volume of 1M NaOH to the collected sputum, leave it at room temperature for 30 minutes, shake it every 10 minutes, centrifuge at 12,000 rpm for 5 minutes, discard the supernatant, add 0.5 mL of saline, and mix. Centrifuge at 12,000 rpm for 5 minutes, discard 400 μL of supernatant, save 100 μL of supernatant, and mix for 15 seconds.

Add 200 μL lysis solution and 20 μL digestion solution, shake and mix, and bath at 56°C for 10 minutes.
Add 500 μL of pellet and mix gently by inverting. If there is a translucent suspension, it will not affect DNA extraction and subsequent experiments.
Place the adsorption column in the collection tube, transfer the solution to the adsorption column, let it stand for 2 min, centrifuge at 12000 rpm at 4°C for 1 min, and discard the waste liquid in the collection tube.
Put the adsorption column back into the collection tube, add 500 μL of washing solution to the adsorption column, centrifuge at 12000 rpm at 4°C for 4 minutes, and discard the waste liquid in the collection tube.
Return the column to the collection tube and centrifuge at 12,000 rpm for 4 minutes at 4°C to remove residual washing liquid.
9. Remove the adsorption column, put it in a new 1.5 mL centrifuge tube, add 30-50 μL of eluent, let it stand for 3 minutes, and centrifuge at 12,000 rpm at 2°C for 4 minutes to collect the DNA solution. The extracted DNA can be used in the next experiment or -20 °C.

Precaution:

The sediment and washing liquid contain irritating chemicals. Please take protective measures during operation to avoid direct contact with the skin and prevent inhalation into the mouth and nose. If it gets into the skin or eyes accidentally, please rinse with water or saline immediately. Seek medical attention if necessary.
It is normal for the lysate to precipitate as white flocs, which can be dissolved in a 37°C water bath.
Storage, transportation and expiry date
The kit can be transported at room temperature and stored at room temperature (15-25°C). The validity period is 12 months. It can be stored in a digestion tank at -20°C to avoid repeated freezing and thawing.
